Rosanne Guijt

Dr Rosanne Guijt is a senior lecturer in the Pharmacy division in the School of Medicine. Her research identifies new solutions at the disciplinary interface between health sciences, chemistry and engineering through the development of new, miniaturised instrumentation. Her current research targets applications in personalised medicine, bioprocess and environmental monitoring and to provide new experimental platforms for example in neuroscience. Her interests range from fundamental fluid dynamics to inform chip design and the development of separation and detection methods to application areas including therapeutic drug monitoring and neuroscience. Her research is conducted in close collaboration with ACROSS.

Biography

Dr Rosanne Guijt completed her undergraduate degree in Biopharmaceutical Sciences at Leiden University, the Netherlands, and her PhD degree from Delft University of Technology where she worked between the Kluyver Institute for Biotechnology and the Institute de Microtechnique (IMT) at the Université de Neuchâtel in Switzerland. Her research involved the development of miniaturized total analytical systems (µTAS) for real-time monitoring of fermentations, and focused on the development of devices for capillary electrophoresis with integrated conductivity detection. She was awarded a fellowship from the Dutch Science and Technology Foundation STW to initiate Lab on a Chip research at the University of Tasmania (µTAS, University of Tasmania). In Australia, she received a 4 year postdoctoral fellowship from the Australian Research Council to work in the Australian Centre for Research on Separation Science (ACROSS), and her research focus broadened to also include the development of flow-through microreactors. Her research strength is in the development of portable and field deployable analytical instrumentation, with applications in counterterrorism and environmental and bioprocess monitoring. She has also explored the use of 3D printing in microfluidics. Following her appointment in the School of Medicine focusing on the development of new technologies for point of care diagnostics and therapeutic drug monitoring. Currently, she resides at the Korean Institute for Science and Technology – Europe in Saarbrucken on an Alexander von Humboldt Fellowship for Experienced Researchers. She has published 67 articles in peer-reviewed journals with an average impact factor over 4.

Research Themes

Rosanne's research aligns o the University's research theme Data, Knowledge and Decisions, developing creative and cross disciplinary solutions to better understand our sparsely observed world. The main focus of this work is in Better Health, on enabling a more personalised approach in medicine, developing a new generation of instrumentation enabling a more widespread use of therapeutic drug monitoring to enable personalised dosage to optimise care ('better treatment'). This technology also provides new platforms for diagnostics, enabling the diagnosis of the onset of disease based on small molecule biomarkers. She also provides new tools that allow answering pathophysiological questions surrounding traumatic brain injury and neuronal communication to inform the design of prevention strategies and early intervention options to stop the chronic disease 'cascade'.

Collaboration

Rosanne is currently involved in an international collaboration with KIST Europe in the general area of miniaturisation of electrophoretic separation techniques. At KIST Europe, Saarbrücken, Germany, she has worked with Prof Manz, prof Abelman, Mr Park and Mr Li, and collaborated with Prof Hankemeier (Leiden University), Prof Synal (ETH Zurich) and Prof Titz (Helmholtz Institute for Pharmaceutical Research). She has initiated links with Prof Arzt (Leibnitz Institute for New Materials) and Prof Lehr (Helmholtz Institute for Pharmaceutical Research). She has also worked on the simulation of conductivity detectors and electrophoretic processes with Prof Ivory (Washington State University) and co-supervised a student with Prof Foret (Institute of Analytical Chemistry, Brno, Czech Republic). She was hosted for 4 weeks in Prof Hauser's laboratory at Basel University, Switzerland in 2009 and 2010.

She has worked closely with the Engineering company Grey Innovation on the development of portable instrumentation for environmental monitoring, and has applied for funding with Grey Innovation and Prof Roberts (UQ) to develop new technologies in the area of Therapeutic Drug Monitoring. She has also worked with First and Bailey at Tasmanian Alkaloids, and Dr Lacher (Pfizer, USA), and collaborates with Dr Crew (CSIRO).

Within the University of Tasmania, she leads the Chippers and CE-ers grouping in the Australian Centre for Research on Separation Science with Prof Breadmore. She has also collaborated with other ACROSS researchers Profs Paull, Hilder, Haddad, Macka, Nesterenko and Assoc Prof. Shellie and Quirino and is part of the ASTECH, the ARC-funded training centre with Trajan. She has co-supervised students with Dr Powell (TIA), Prof Shaballa and Dr Makwana (Agricultural Science), Dr Henderson (Engineering), Prof Bolch (AMC), Assoc Prof Dickson (Menzies) and Dr King (School of Medicine). Within Pharmacy she has worked with Assoc Profs Güven and Ferguson and Drs Jacobson and Westbury. She has worked with Dr Howells (Education) studying the impact of gratitude in RHD supervision and led a research project investigating strategies for successful returns from parental leave with Dr Hardy (Economics), Prof McDonald (Law), Assoc Prof Jones (Zoology), Dr Green (IMAS), Assoc Prof Leanne (Arts), Ms James (AMC), Prof Dickinson (Menzies), Dr Stephenson (Education) and Dr Martin (Business).

Current projects

Rosanne's research focuses around the development of portable and field deployable instrumentation based on electrophoretic separation processes, targeting applications in health sciences, food safety and environmental and bioprocess monitoring. This includes the development of applications in therapeutic drug monitoring based on the electrokinetic size mobility trap they recently invented. Process and environmental monitoring systems are being developed based on the sequential injection capillary electrophoresis technique they developed, enabling for automated sample injection using small sample volumes over extended periods of time. She also has an interest in the development of microfluidic devices for neuronal cell culture, currently focusing on enhancing the understanding of the pathology in traumatic brain injury and the role of glia in neuronal communication. Rosanne is interested in developing additional cell culture-based microdevices to expand her research into systems pharmacology.

Microfluidic technology to help understand physical damage to brain cells (2015 - 2017)$415,500
Description
Understanding the organisation, structure and mechanisms of the human brain and nervous system remains one of the biggest challenges of science. This project will develop a new cell culture platform to form defined molecular networks of brain cells and to monitor changes throughout the network in response to a small localised injury within the network. This innovative platform will be used to help understand changes within cells in response to physical damage to networks of brain cells. This is one of the major causes of death and disability in developed nations, and is identified as a risk factor for a range of neurodegenerative diseases including Alzheimer's, Parkinson's and motor neuron disease.

Neuroprotective function of novel short chain-quinones (2013)$9,090
Description
Most eye disorders are associated with mitochondrial dysfunction. Based on the mitochondria-protective drug idebenone, we will synthesize novel, improved neuroprotective compounds. These compounds will be tested in a microfluidics cell culture model of mechanical stress-induced neuro-degeneration to identify new treatments strategies against retinal degeneration.

Description
A Lab on a Chip for Na+ measurements in plants is proposed, offering a long overdue solution to the lack of appropriate techniques to study the mechanisms of Na+ uptake, transport and compartmentation. Na+ transport is a key determinant of salt tolerance, but a good understanding of its transport mechanisms is lacking since no appropriate measurements tools are available. Using the system proposed here, Na+ is separated from interfering ions, allowing fast and selective measurements. A series of studies towards Na+ uptake, transport and compartmentation in plants will be conducted using this Lab on a Chip.

Research Supervisions

Rosanne is a dedicated and caring supervisor of a mainly international research team with 100% completion rate; she graduated 6 PhD students. As Rosanne's research is multidisciplinary, her research team has included pharmacists, chemists, engineers and biotechnology specialists. Creativity and cross disciplinary interaction are quintessential for high quality, high impact research, and Rosanne aims to create an environment where creativity can evoke. To inform her supervision practice, Rosanne initiated a study towards the impact of gratitude practice on the student supervisor relationship and found this improves deeper thinking whilst enhancing the students and supervisors overall sense of well-being.

All of Dr Guijt's research projects cross disciplinary boundaries and involve at least 1 other supervisor, with the importance of access to ACROSS laboratories, infrastructure, know-how, reputation and critical mass, this person is likely to be a member of ACROSS.
