Inflammation is the process by which the body recognises an infection or tissue damage and actively recruits cells and initiates processes of the immune system to respond to it. Inflammation is usually the first step in the body's immune response. Its most recognisable early manifestations are redness at the site of infection or swelling around an injury.

Professor Heinrich Korner's research investigates the steps in the body’s inflammatory response, the molecular agents involved, and what happens when this process goes wrong.

Inflammation is a vital part of the normal immune response and it is central to the process by which the body heals itself. It also generates immunity to future incursions of disease agents.

However, chronic inflammation – when inflammation is triggered unnecessarily, or when it goes on too long – can lead to auto-immunity. In this condition, the cells of the immune system and the toxic chemicals they produce, which normally fight off infectious microbes, instead turn on the body’s own tissues.

'No one really knows what causes auto-immune diseases; causality has not been conclusively established,' Professor Korner says.

Rheumatoid arthritis is a good example of an auto-immune condition linked to chronic inflammation. Multiple sclerosis (MS) is another autoimmune disease which affects the central nervous system.

Professor Korner has studied the role of tumour necrosis factor (TNF), one of a group of molecules called cytokines, which coordinates the normal process of inflammation. His research demonstrated that TNF plays a key role in the process by which a class of immune cells known as macrophages switches to a pro-inflammation response. In mice in which the gene for TNF has been deactivated (TNF-knockout mice), a large proportion of the macrophages do not make this shift to the pro-inflammation state. As a consequence, the immune response is far less effective, particularly against intra-cellular pathogens.

Over-expression of TNF may, however, be one of the triggering events for autoimmunity in conditions like rheumatoid arthritis. A new class of drugs has been developed to suppress TNF and these drugs have been successful in suppressing rheumatoid arthritis. What is less well understood, however, is how anti-TNF therapy might compromise the immune system’s ability to perform its normal function, similar to the situation in the TNF-knockout mice.

Professor Korner is also carrying out research into the link between multiple sclerosis and vitamin D. In MS, the mechanism by which the immune system is triggered to attack the cells of the body's own nervous system remains a mystery. However, it is assumed to be associated with a deficiency of vitamin D. Professor Korner has shown that vitamin D acts as an anti-inflammatory agent, at least in tissue cultures. However, he is cautious about suggesting that vitamin D could be used to treat or prevent MS.

'MS is a complex disease and these results have only been shown in vitro. There is a lot we don't know about what goes on in a real person and how genetic, environmental and other factors all interact.'

Professor Korner is also collaborating in research investigating another group of cytokines, known as interleukins. Specifically, this research has shown that in mice lacking a certain kind of inflammasome – a large protein complex that is involved in the activation and secretion of interleukin – inflammation in the bowel can develop into bowel cancer.

'The link between inflammation and cancer is unexpected,' he says. 'This research is at an early stage and much more work needs to be done to understand the underlying connection.'

Biography

Before joining the University of Tasmania, Heinrich studied Biology at the University of Wuerzburg, Germany, and completed his PhD thesis at the Institute for Virology and Immunology. After completing a post doctoral period at the Max-Planck-Institute in Freiburg, he moved to Australia and established gene-deficient mouse strains in the laboratory of Dr Jon Sedgwick at the Centenary Institute in Sydney. In 1998 he went back to Germany to establish his own research laboratory. In 2003, he left again for Australia after being appointed to a tenured position at James Cook University, Townsville Australia. In 2011 he relocated to the University of Tasmania following his appointment as New Star Professor at the Menzies Institute for Medical Research.

Research Themes

Heinrich's research aligns to the University's research theme of Better Health. His research is interested in inflammation in general and he uses different models of infection and autoimmunity to better understand the contributions of the different components of the immune system.

Collaboration

Prof Korner has excellent research connections nationally as well as internationally. He collaborates with Prof Robert Brink (Garvan Institute, Sydney), Assoc Prof Matthew Cook and Prof Carola Vinuesa (ANU, Canberra) a well as Assoc Prof Dave Tarlinton (WEHI, Melbourne) to investigate the role of chemokine receptors. Furthermore, he has collaborations with Assoc Prof Matthew Sweet (IMB, Brisbane) and Dr. Antje Blumenthal (Diamantina, Brisbane) and Prof. Christian Bogdan (Erlangen, Germany) on his project examining the role of TNF in macrophage maturation. Finally, he works with Dr David Sacks (NIH, USA), Prof Mark Jenkins (University of Minesota, USA), Prof. Dale Godfrey and Assoc Prof Chris Engwerda on a project to develop a better way to study the memory response to L. major which will be important for vaccine development. Many of these collaborations have resulted in joint publications.

Prof Korner's work has been supported by funding through the NHMRC in addition to a number of different smaller grants from a broad range of sources. In 2011 he has been successful to receive a Ramaciotti equipment grant for the purchase of an analytical 9-colour flow cytometer (Cyan, Beckman Coulter) and a Miltenyi AutoMax. He has been appointed as one of 5 New Star Professors at the University of Tasmania. This position is fully covered by a central grant of the University of Tasmania until the end of 2015.

Functional consequences of Multiple Sclerosis-associated genetic changes in Vitamin D metabolism genes in T cells and the pathogenesis of autoimmunity (2014 - 2015)$24,950
Description
It is now well recognised that the risk of developing Multiple Sclerosis (MS) is governed by a complex interplay between genetic and environmental factors. Genes within the vitamin D pathway have been implicated in MS risk as has lower levels of vitamin D. How these 2 factors interact to alter immunity and increase the risk of developing autoimmune disease is not clear. This project looks at the functional consequences of a change in the genetic code in known MS risk genes that are in the vitamin D metabolic pathway and how these changes alter the function of T lymphocytes, the immune cells thought to play a major role in the inflammation that is characteristic of MS.
