Greg Woods is a Professor of immunology at the School of Medicine and a Principal Research Fellow at the Menzies Institute for Medical Research. He teaches immunology and his research involves the immunology of Devil Facial Tumour Disease (DFTD). In 2011, as part of a five-member team, he was awarded the Sherman Eureka Award for Environmental Science and received a Fellowship of the Faculty of Science, Royal College of Pathologists of Australasia; and in 2015 received The Australian Society for Medical Research Certificate in Recognition of Distinguished Service to Medicine, Science and Community.
Biography
Professor Greg Woods completed a BSc (Hons) undergraduate degree in 1978, at Monash University and completed postgraduate (PhD) degree at the University of Tasmania, in 1984. In 1985 worked as a junior research fellow at the Department of Medicine, University of Tasmania. For the next two years he was at the Mt. Sinai Hospital Research institute, Toronto Canada as postdoctoral fellow. In 1989 he obtained a lectureship at the University of Tasmania and was promoted to professor in January 2013. In 1993 he was a visiting scientist at the Clinical Research Centre, Northwick Park Hospital, Harrow, England and in 2003 a visiting scientist at the University of Edinburgh, Edinburgh, Scotland.

Prof Woods teaches within a range of undergraduate units offered by the School of Medicine with a focus on immunology but also in medical microbiology, pathology and molecular biology. Greg's versatile teaching ability has enabled him to engage students from a range of backgrounds including science, medicine and pharmacy students. His successful use of analogies and visual aids to assist in the explanation of complex scientific ideas has made him a popular lecturer.

Research Themes
Greg's research aligns with Environment, Resources and Sustainability. Some environmental stressors require immediate solutions to prevent extinction. Emerging infectious diseases, like the Devil Facial Tumour Disease (DFTD), must be managed while infectious zoonotic diseases threaten both biodiversity and human populations. Since commencing research on devil facial tumour disease in 2006 when this disease was first identified as a serious threat to the survival of the Tasmanian devil Greg has been leading the immunology program. With limited and basic tools to analyse the immune response Greg's laboratory has been able to clearly demonstrate that the Tasmanian devil has a robust immune response. Collaboration has been an important aspect in understanding DFTD and the collaborations have resulted in significant advances including the understanding that Tasmanian devils have a limited MHC-diversity and that DFTD derived from a Schwann cell. This latter observation has led to the use of periaxin as an accurate and effective diagnostic marker of DFTD.
The current research emphasis is to understand the devil's immune response to DFTD with the ultimate objective of a vaccine. The research has now provided important advances, one of the most significant is that immunised devils can produce an immune response to DFTD. The challenge is to translate this into a vaccine that can be used to protect wild devils.

- The current DFTD vaccine can induce anti-DFTD immune responses, but the scalability and efficacy of the vaccine needs to be improved to deliver a broad conservation impact. - This proposal seeks support for an advanced, multi-purpose mass spectrometry platform for high-throughput and targeted biomolecular analysis, including proteomics and metabolomics. The intended purpose of the proposed infrastructure is to provide a centralised state-of-the-art facility that supports internationally competitive research programs in plant science, agricultural research, food safety, animal and human health research and separation science at the University of Tasmania (UTAS) and within the broader Tasmanian scientific community. - We have developed a system that allows us to switch genes on/off in devil facial tumour (DFT) cells. The DFT-Off cells have the potential to be used as a live-attenuated vaccine, which generally perform better than killed vaccines, that provides long-term protection against both forms of DFT disease (DFTD).

- Immunotherapeutics targeting PD-1 and CTLA4 have shown unprecedented success in treating human cancers. Surprisingly, little is known to date on the potential for translating these human immunotherapy approaches into cancer treatments for veterinary medicine. The Tasmanian devil facial tumours (DFTs) and canine transmissible venereal tumour (CTVT) provide unique opportunities to assess immunotherapeutic treatment regimens for two naturally occurring tumours. Our ongoing work on the DFT has already developed ten anti-PD-1, nine anti-PD-L1 (aka B7-H1), and four anti-CTLA4 monoclonal antibodies (mAbs) that are highly specific for devil proteins. We have developed a system that can be used to rapidly generate new mAbs against additional target proteins. Importantly, this system can be readily applied to the production of antibodies that target proteins in other species. We have already begun cloning target genes in dogs, and will begin production of recombinant dog proteins, cell lines, and mAbs in 2016. The devil immunology group will develop dog-specific mAbs, and perform in vitro testing functional testing of the mAbs. The first batch of antibodies for PETization will be delivered to Nexvet in January, 2017. The initial functional testing of the antibodies performed by the devil immunology team will generate vital preliminary data to support planned ARC Linkage Project application in 2016-2017 in order to leverage funds for ongoing collaborative research. Establishing a solid connection between Nexvet and the devil immunology team prior to submission of the ARC Linkage Project application will greatly increase the probability receiving a Linkage Project award and leveraging funds. This research could help save an iconic Australian animal from extinction, develop immunotherapeutics to improve and extend the lives of pets around the world, and help to drive an emerging market for pharmaceuticals for pets.

- Monitoring the immune response of vaccinated Tasmanian devils requires analysis of cell mediated immunity .The current assays we use demonstrate killing of DFTD cells but fail to reveal cytotoxic cell activation and themeans of cell death. We plan incorporate a new assay that employs fluorescent in situ hybridisation andbranched DNA amplification technique for simultaneous measurement of RNA transcripts in single cells usingflow cytometry. The advantages of the proposed assay is that it provides a rapid and biologically relevantanalysis of fresh blood samples taken from Tasmanian devils and measures the activity level of these cellsequivalent to when they were in circulation. This will determine if cytotoxic cells are activated to induce targetcell death. We will be able to analyse key gene expression markers for activation such as IFN-gamma andLAMP-I.

This project aims to discover if re-expression of MHC molecules allows DFTD cells to be recognised as an allograft and ultimately be killed by the devil immune system. The demonstration that tumour cells can be recognized by cytotoxic T cells will aid the design of immune based strategies for prevention and therapy.
